DBCO (Dibenzocyclooctyne) is a cycloalkyne that can reacts with azides via strain-promoted 1,3-dipolar cycloaddition in aqueous solution, a bioorthogonal reaction also termed Cu-free click reaction.  This reaction has excellent selectivity and biocompatibility, such that the complimentary reagents can form covalent bonds with richly functionalized biological systems. The Cu-free click reaction has been a powerful tool in catalyst-free bioconjugation. DBCO reagents posses fast kinetcs and stability in aqueous buffer and they can be used to label azide-modified biomolecules with high specificity and reactivity.

Copper-free Click Reaction Procedures:

Recommendation: Add 1 mole equivalent of limiting reagent to 1.5-3.0 mole equivalents of highest abundance reagent. Incubate the reaction at room temperature for 2-4 hours or 2-12 hours at 4 degree celcius. The reaction is now ready for purification.

ITO (Indium Tin Oxide) and FTO coated glass slides provided from Nanocs were made by coating ITO or FTO to one side of soda lime glass. These ITO and/or FTO coated glass, also called transparent conductive glass (TCO), come with different surface resistivity from 5 ohm/sq to 100 ohm/sq, with thickness from 0.5 mm to 2.2 mm. ITO and FTO coated glass has been used in LCD/OLED devices and many other devices requiring high electrical conductivity and good optical transmission. Our ITO/FTO coated glass has excellent surface roughness around +-5 nm. Thickness of our ITO or FTO coating ranges from 50 nm to 700 nm depends on their surface resistivity. Optical transmission of these ITO coated glass is over 85% in the visible range.

Nanocs SnapLinkTM pegylated succinimidyl cyclohexane Maleimide (SMCC) amine is a heterobifunctional crosslinker that can react with sulfhydryl and amien reactive groups, such as NHS ester. This crosslinker contains a sulfhydryl reactive maleimide group and a primary amine group bridged with PEG arm. The hydrophilic polyethylene glycol (PEG) spacer arm imparts water solubility that is transferred to the crosslinked molecules , thus reducing aggregation of labeled proteins in solution. The PEG spacer arm also gives the cyclohexane maleimide PEG amine a long and flexible structure to minimize steric hindrance for its reaction to other molecues. Compared to other maleimide NHS ester, SMCC has higher stability in aqueous solution because of its cyclohexane bridge. Maleimide group reacts with available thiol or sulfhydryl at pH 6.5~7.5 to form stable thioether bonds.
