胰蛋白酶、北京销售胰蛋白酶

胰蛋白酶消化液一共有三种，分为胰蛋白酶－EDTA消化液（不含酚红）；胰蛋白酶－EDTA消化液（含酚红）；胰蛋白酶消化液（不含酚红），可根据客户的需要来选择。
其中，胰蛋白酶含量为0.25%（2.5g/L），EDTA含量为0.02%（0.2g/L）。PBS为常用的不含钙镁的细胞培养用PBS。经无菌过滤，可直接使用。

胰蛋白酶使用前，可适当分装一下，使用时，是预热到37度，镜下控制消化时间。

 

产品简介：

胰蛋白酶消化液一共有三种，分为胰蛋白酶－EDTA消化液（不含酚红）；胰蛋白酶－EDTA消化液（含酚红）；胰蛋白酶消化液（不含酚红），可根据客户的需要来选择。

其中，胰蛋白酶含量为0.25%（2.5g/L），EDTA含量为0.02%（0.2g/L）。PBS为常用的不含钙镁的细胞培养用PBS。经无菌过滤，可直接使用。

 

保存条件：

-20℃保存，一年有效。客户收到产品后，可以适当的分装后冻存，每次取一支出来使用。

 

使用说明：

1. 贴壁细胞的消化：

a） 吸去培养液，用无菌的PBS、Hanks液或无血清培养液洗涤细胞一次，以去除残余的血清。

b） 加入少量消化液，略盖过细胞即可，室温放置30秒至2分钟或者更长时间。不同的细胞消化时间有所不同，必要时可放37℃消化。

c） 显微镜下观察，细胞明显收缩，并且肉眼观察培养器皿底部发现细胞的形态发生明显的变化；或者用枪吹打细胞发现细胞刚好可以被吹打下来。此时吸除胰酶细胞消化液。加入含血清的*细胞培养液，吹打下细胞，即可直接用于后续实验。

d） 如果发现消化不足，则加入胰酶细胞消化液重新消化。

e） 如果发现细胞消化时间过长，未及吹打细胞，细胞已经有部分直接从培养器皿底部脱落，直接用胰酶细胞培养液把细胞全部吹打下来。1000-2000g离心1分钟，沉淀细胞，尽量去除胰酶细胞消化液后，加入含血清的*培养液重新悬浮细胞，即可用于后续实验。

 

2. 组织的消化：

不同的组织需要消化的时间相差很大，通常以消化后可以充分打散组织为宜。

 

 

注意事项：

1.在分装胰蛋白酶-DETA消化液的过程中要特别注意避免消化液被细菌污染。

2.胰蛋白酶-DETA消化液消化细胞时间不宜过长，否则细胞铺板后生长状况会较差。
